A web page for each user which links to all the playlists that the user has created
Smart playlists: playlists based on criteria and which dynamically update. These are really just glorified saved searches but it means I could set up a search for artists that aren't yet in Spotify and get notified when they turn up.
Support for libre.fm as well as last.fm
A web-based API so that other people could improve the service with things like automated recommendations.
Make use of MusicBrainz. Using or just cross-referencing the MusicBrainz identifiers would make it a lot easier to find music. It could be as simple as a link from the Spotify web page for a track to the MusicBrainz page for that track.

It was a pretty good event given: an audience consisting mostly of .Net developers, a terrible venue (no 3g in the main hall and the audience sits in darkness which means the speakers can't build rapport as they can't see anyone out there) and it's a single track conference so there that isn't much interaction or content.
I tend to prefer multi-track events with plenty of hallway time.
